Principle
This method is based on the conversion calcium salt in the salt of
another metal ( Silver ) which itself is opaque..
Solutions
A) Citrate buffer pH- 4.5
0.2M Disodium hydrogen phosphate - 283
gms
Distilled Water - 100ml
B ) 0.1M Citric acid
Citric
acid - 2.1 gm
Distilled
wate - 100ml
Working solution
Solution A - 9.09ml
Solution B - 10.9
5%, Silver nitrate
Silver nitrate - 5
gms
Distilled water - 100ml

1% Eosin.
Procedure
1) Deparaffinise two sections in xylene for 10mnts
2) Bring the section to water through descending series of alcohol
.
3) Immerse one section in citrate buffer for 20mnts.
4) Wash both the slides well in distilled Water
5) Flood slides with silver nitrate.
6), Expose to bright sunlight or ultraviolet light for 30-45
minutes on to a 60 Watl electric bulb at a range of 4-5 inches for 30-60
minutes.
7) Wash in several
changes of distilled water
8) Treat
with 5% solution thiosulphate for 2-3 mnts
9) Counterstain with eosin
for 30 Secs
10) Wash in water,
dehydrate, clear and mount.
Result
Calcium deposits - Black
Background - Pink
