VANGEISON STAIN
Principle
The method depends upon thę of tissue and the size of the dye
molecule . Thus collagen Whish is relatively permeable takes red colour with acid fuchsin
and muscle which in far less permeable takes yellow with picric acid . Weigert’s
haematoxylin which is mordanted to an iron salt
avidity to withstand treatment with
picric acid . which effectively removes alum haematoxylin from nclei.
Solutions
1.
Weigert’s haematoxylin
Solution A
Haematoxylin - 1gm
Absolute Alcohol - 100ml
Solution B
30% Aqueous ferric chloride - 4ml
Concentrated Hydrochloric acid - 1ml
Distilled water - 95 ml
Mix equal quantities of A and B before use. The solution
should be dark black in color .
2. Vangeison
solution ( Red Color )
Saturated
aqueous picric acid - 100ml
1% Acid Fuchsin
Procedure
1. Reparaffinise the section in Xylene for 5-10
mnts
2. Bring the section it alè ugh descending
series of alcohol
3. Stain with Haematoxylin for 5-10 mts
4. Wash it in water
5. Differentiate it in 1% acid alcohol
6. Keep it in running tap water for 5-10 mnts
7. Stain with vangeison Solution for 2 mts.
8. Drain the solution and blot with filter paper
9. Picric acid will be
dissolved when washed in water
10. Clear and Mount.
Result
Collagen - Red
Nuclei - Blue
Muscle - Yellow
