MUCCARMINE
Principle
Mucicarmine has got specific size. The aluminium salts in the
solution form a chelate compound with carmine, thus conferring a net positive charge on
the molecule and consequent binding to the tissue polyanion.
Solutions
1)
Mucicarmine solution
Carmine - l gm
Anhydrous
aluminium chloride - 0.5gm
50% alcohol - 100ml
Take 2 ml distilled water in a test tube, add carmine and
aluminium chloride. I leat it under flame, until the solution becomes syrupy.
Dissolve the solution by adding 50% alcohol little by little. Cool, filter and
store it at 4°C.
2)
Weigerts Iron haematoxylin
Solution A
Heamatoxylin - 1
gm
Absolute alcohol - 100ml
Solution B
30%
Aqueous ferric chloride - 4ml
Concentrated
hydrochloric acid - 1 ml
Distilled water - 95
ml
Mix equal amount of A and B before use.
3)
Metanit Yellow:- 0.25%
Metanil
Yellow - 0.25 gm
Distilled
water - 100 ml
Procedure
1)
Deparaffinise the section in xylene for 10 mts
2)
Bring the section to water through descending series of alcohol.
3) Stain with iron Haematoxylin for 5-10 mts.
4) Wash it in water. Differentiate in 1% acid alcohol.
5) Keep it in running tap water for 5-10 mts
6) Stain with mucicarmine solution for 45-60 mts. . (Dilute 1 part of
stock solution with 3 parts of tap water).
7)
Wash it in distilled water. Counter stain with metanil yellow for
30 sec.
8)
Wash it in water, dehydrate, clear and mount.
Result
Mucin - Pink
Nuclei - Black
Background - Yellow.
