Frozen
section is a very useful technique which is used
1) For the demonstration of fats, lipids
2) For the rapid
preparation of sections for diagnostic purposes.
For cutting sections, rotary microtome kept inside a chamber, the temperature
of which is around -20°C is used. The temperature can be adjusted according to
the nature of the tissue
.
Procedure
1) Keep the unfixed tissue on the top of the holder.
2) Add either water or the
cryo embed media around the tissue.
3) Wait for the tissue to
get frozen.
4) Fix the holder to the
microtone and bring the knife close to the tissue
5) Trim the tissue.
6) Clean the surface of the knife with the help of a brush.
7) Adjust the anti-roll, such that edge of the anti-roll and the
edge of the knife are parallel.
8) Reduce the thickness to 5 µ and slowly cut sections.
9) Take the section which is between the knife and the anti-roll
on a plain slide.
10) Slightly heat the slide.
11) Dip it in water and keep in heamatoxylin for 2 minutes.
12) Wash it in water and slightly differentiate it with 1% acid
alcohol
13) Keep it in running tap water for bluing, even Saturated
lithium carbonate is used so quick blueing
14) Stain with eosin for 10.15 seconds.
15) Wash in water, dehydrate, clear and mount.
