                                FOUCHET'S STAN

Principle

The bile pigment is converted to the green colour of biliverdin and blue cholecyanin by the oxidative action of the ferric chloride in the presence of trichloroacetic acid.


Solutions

1)  Fouchet's solution
                        25% Aqueous trichloroacetic acid              -           25 ml
                         10% Aqueous ferric chloride                      -           10ml

          Freshly prepared before use.
2)  Vangeison Solution
                   Saturated solution of picric acid  -        100ml
                   1% Acid fuchsin                                  -        10ml










Procedure

1)   Deparaffinise the section in xylene for 10mts.
2)   Bring the section to water through descending series of alcohol.
3)   Rinse with distilled water.
4)   Treat with Fouchet's solution for 5 minute.
5)   Wash well in running tap water for 10 minute.
6)   Rinse in distilled Water.
7)   Counterstain with Vangieson solution for 1 min.
8)      Blot with filter paper, clear and mount.



Result

          Bile pigments                   -        Emerald to bluegreen.
          Muscle                            -       Yellow

          Collagen                          -        Red.
