Principle
Melanin have the ability to reduce solution of ammoniacal silver
nitrate to metallic silver.
Solutions
1)
Ammoniacal silver solution:
To 10%
aqueous silver nitrate add strong ammonia. A precipitate will be formed. Add ammonia drop by drop until the
precipitate dissolves. Add 10% silver nitrate drop by drop, until a muddy color
is formed. Dilute with equal amount of distilled
water. Keep it in dark overnight.
2) 1% Sodium thiosulphate.
Sodium thiosulphate - 1gm
Distilled water - 100ml
3) 1% Eosin.
Eosin - 1 gm
Distilled
water - 100ml
Procedure
1) Deparaffinise the section in Xylene for 10 mts.
2) Bring the section to water through descending series Of
alcohol.
3) Rinse the section
thoroughly in distilled water.
4) Keep the section in the beaker containing the ammoniacal silver
solution at 60°C for 45-60 mts (Until light brown colour develops).
5) Wash it in distilled water.
6) Treat with sodium
thiosulphate for 2-3 mts.
7) Wash it in water.
8) Counterstain with eosin for 1 mt
9) Wash it in water, dehydrate, clear and mount
