MUSEUM TECHNIQUES
The specimen to be mounted is treated with the following
solutions. +
KAISERLING SOLUTION I
(For fixing the tissue)
It contains
a) Formalin - 400ml
b) Potassium nitrate-30gms
c) Potassium acetate-60gms
d) Tap water
(filtered)-2000ml (2L)
Specimens can be immersed for six months in Solution I alone.
KAISERLING SOLUTION II (For restoring the colour)
It contains 80% (v/V) ethyl alcohol. It is used to restore color
of the specimen in an emergency.
KAISERING SOLUTION III (A mounting fluid)
It contains
a) Glycerin - 300ml
b) Sodium acetate - 100gms
c) Formalin - 5ml
d) Tap water - 1000ml
(1 liter)
There are two types of
mounting
1.
Mounting in a glass jar
2.
Mounting in polythene covers
1) Mounting in a glass iar:
Depending upon the size of the specimen, the jar is selected and
appropriate glass rod is selected. It is bent with pressurized flame and made
to fit into the jar,
The
specimen to be exhibited is tied with the help of nylon thread to the glass rod
which has been made and allowed to stand inside the glass jar. Now
it is filled with kaiserling solution III until the entire specimen which was
made to stand inside the jar is immersed. Then add 0.4 g/dl sodium
hydrosulphite immediately before sealing. The jar is covered with appropriate
lid with molten wax permanently to avoid evaporation of mounting fluid.
2) Mounting in a polythene cover:
It is an economical method
and it is easy to exhibit to the students by demonstrating the outer
and the cut surface of the specimen.
The specimen is kept inside the packet and it
is filled with Kaiserling solution III and sealed with a horizontal heat
sealer. Care should be taken not to introduce air bubbles or air space inside
the packet. Then to the top of the mounted specimen which is showing the gross
surface. the cut surface or a slice of anterior surface is introduced and
filled with Kaiserling solution III and sealed with horizontal heat sealer in
the same manner. Now the packet is clearly labeled with biopsy number and the
year with the patients name, age, sex, hospital no. and the clinical diagnosis
with the biopsy report.
Then it is ready for exhibiting in the
museum.
Similarly all the
pathologically important specimens are mounted in either of the method and
cataloguing is done annually and a register is maintained with all the details
regarding the specimen.
