MELANIN BLEACH
MELANIN BLEACH
Melanin is insoluble in almost all organic solvents. Strong
oxidizing agent like potassium permanganate will bleach the melanin pigment.
Solutions
1) 0.25% Potassium permanganate.
Potassium
permanganate - 250mg
Distilled
water -
100ml
2) 1% Oxalic acid
Oxalic acid - 1gm
Distilled water - 100ml
Procedure
1) Deparaffinise the section in xylene for 10 mts.
2) Bring the section to water through descending series of alcohol.
3) Treat with potassium permanganate for 30mts. The time required for
1. bleaching varies according to the site and degree of pigmentation.
4)
Wash it in water.
5)
Treat with oxalic acid for 30mts.
6)
Keep it in running tap water for 10mts.
7)
Do the routine Haematoxylin and eosin stain.
8)
Dehydrate, clear and mount.

Result
Melanin pigment
will not be seen in the section which is bleached.
