GOMORIS
METHANAMINE SILVER
Principle
The carbohydrates in the fungi are converted to dialdehyde by
chromic acid treatment, which impregnate the silver.
Solutions
1)
Gomoris silver methanamine solution
5%,
Silver nitrate - 5 ml
3%
Hexamine in distilled water - 100ml
Add silver nitrate to methanamine solution shaking until the
precipitate which first forms dissolves. Store it in brown bottle at 4°C.
Working solution
Silver methanamine solution
- 25 ml
Distilled water - 25 ml
5% Borax - 3ml
Glasswares should be washed well and rinsed thoroughly in
distilled water. Metal forceps should not come in contact with the solution.
2) Gold chloride
Gold
chloride - 200 mg
Distilled
water – 100 ml
3) 3% sodium thiosulphate
Sodium thiosulphate - 3 gm
Distilled water - l00
ml
4) Light green
light green - 200 ml
0.2% acetic acid - 100ml
Procedure
1. Deparaffinise in Xylene
for 10 mts.
2. Bring the section to water through descending
series of alcohol.
3. Oxidise with chromic
acid for 30 mts
4. Rinse the section
thoroughly in distilled water.
5. Keep the section in a beaker containing the
working solution, inside the incubator at 60°C.
6. Check for the development of light brown
colour, usually it takes around 60 minutes for the development of brown colour.
7. Wash it in distilled
water.
8.
Treat with gold chloride
for 10 mts wash it in water.
9.
Treat with sodium
thiosulphate for 2-3 mts
10.
Wash it in water.
11.
Counter stain with light
green for 30 sec. - 1 minute.
12.
Wash it in water,
dehydrate, clear and mount.
Result
PAS
positive structures, basement membrane, fungi, mucin etc. - Black to Brown
Background
- Green.
