Sections cut vertically (SCORES)
|
-Knife contains small nicks
-Knife edge to be cleaned
-Tissue has calcified areas

Sections cut or roll up
|
-Knife is blunt
- Tilt of knife is too-much ( so reduce clearance angle)
|
|
Sections are alternatively thick and thin
|
Microtome adjusting Screws need-tightening

Sections crumble on cutting

-Knife is blunt
- Wax is too soft and needs ice applied to the
Cutting-Surface
- Wax is crystallized due to slow cooling or contamination with
water or clearing agent

Ribbon of
sections curved
|
-Block-edges-are not-parallel-to-each other
- Block edges are not parallel to knife edge
|
Serial
deeper - Take ribbon sections, trim a little bit and again take ribbon
sections. Like this take 3-4 ribbon sections.
Deeper - If the whole section is not exposed, adjust
the position of the block, so that the unexposed portion is trimmed
first. Trim until the whole the section is exposed and take ribbon sections. If
the whole section is exposed trim a little bit and take ribbon section.
Thinner- Reduce the micron adjustment to 1-2µ and cut slowly and take section as thin as
possible.
Rotate block
Some tissues need orientation e.g.-tubes should embed
perpendicularly. If it is embedded longitudinally melt the block, rotate the tube
by 90* and reembed it. ' Sometimes, the cutting surface will be the reverse
side. Melt the tissue rotate it 80° and reembed.
