CONGO RED
Principle
The staining is based on the selective affinity of the dye Congo
red for amyloid.
Solutions
1)
1% Congo red (orange
colour)
Congo red -
1 gm
Distilled water - 100 ml
2) Haematoxylin,
3) Saturated Lithium
Carbonate.
Procedure
1) Deparaffinise the section in xylene for 10mts.
2) Bring the section to water through descending series of
alcohol.
3) Stain with Congo red for
30 mts.
4) Wash with tap water
.5) Flood slide with saturated aqueous solution of lithium carbonate
for 15 seconds
6) Keep it in running tan water for 5-10 mt
7) Counterstain with Haematoxylin for 1-2 mts
8) Wash it in water
9) Differentiate it in 1% acid alcohol.
10) Keep it in running tap
water for 5-10 mts
11) Dehydrate, clear and mount
Result -
Amyloid - Pink
to red.
Nuclei - Blue
