RED
Principle
This method is based on the ability of certain dyes (alizarin red)
to form lakes with calcium salts.
Solutions
Alizarin
red - 1gms
Distilled
water - 50ml
Add 0.5% ammonia until pH 4.2 is reached, using a pH meter. The
solution should be deep iodine in colour.
Procedure
1) Deparaffinise the section in xylene for 10mts.
2) Bring the section to water through descending series of
alcohol.
3) Place the section in a
coplin jar filled with alizarin solution for 1-5 minutes.
4) Blot and rinse in
acetone for 30 seconds.
5) Treat with equal parts of acetone Xylene for 15 seconds.
6) Rinse in Xylene and
mount.

Calcium deposits - Orange red
